Liquid Crystalline DNA
In the laboratory scientists usually investigate the physical and biological properties of DNA in dilute solution. However, DNA exists in vivo in domains where the localized concentrations are very high. As the aqueous solution concentration of DNA is slowly increased, the macromolecule undergoes spontaneous phase transitions to form at least three distinct liquid crystalline phases which are termed lyotropic phase transitions in the liquid crystal field. The digital images presented below capture short rod-like molecules of DNA that form a high-density liquid crystalline columnar hexatic phase.

Polarized light images were captured using an Intel Play QX3 computer microscope modified for crossed polarized illumination. Additional light was applied to the specimens using light pipes from a Dolan-Jenner Model 180 fiber optics illuminator containing a 150 watt projection lamp. A 5/16-inch hole was drilled into the light mixing chamber at the base of the microscope and the light pipe was pointed towards the far wall of the chamber to increase illumination.
